Are you able to actually see the screw rotating when you turn counterclockwise? Is it possible that you over-tightened the screw, damaging the threaded insert?

The screw is not supposed to come out all the way. Loosen it a little bit then slide the back downwards a little bit. If the back does not slide down, loosen it a little more until the back slides down and off.

I am assuming you are trying to open the doorbell up for the first time so that you can install the batteries and set the switch. If you have already mounted it and forgot how to separate the two sections, you would loosen the sctew a little and try to push up on the front piece with the black face. If you can't push it up, loosen the screw a little more until you can push it up and off.
