I have a USB stick plugged into the rear USB port of the router. The USB LED (#2) is lit. How would I modify and save a password database contained on the USB stick from my desktop computer (which is cabled to the router)?

If your referring to the Pro base USB, you can't. There is no access into the base

Well, I don't know what the Pro base USB is so I can't relate to your answer. The USB stick is just a plain "thumb drive" plugged into the back of the router. I need to be able to access this as if it were plugged directly into my computer.

None of the Arlo products that support local storage allows access to that storage without physically removing it from the device and connecting it to the computer. IOW, it won't act as a NAS.
